"Scout" would be an obvious choice, as they know location of stairs.
And what do yo think as the main obstacle of nightmare Ao100/ArAo666?
That is ammo supply. Number of horde of monsters is crazy, and,
common boss monsters (bruisers, shamblers, cyberdemons, lava elementals) have extremely huge HP than other difficulties.
So, the success of N! Ao100/ArAo666 depends on
"how early you can assemble nanomachic weapon(+BBBN, Whizkid 2)".
nanomachic laser rifle+F is the best, nano super shotgun+P is second best (this is not enough if you aren't going with Shottyhead build),
nanomachic plasma rifle/minigun+F would be worse alternatives.
Anyway, that is your must-have item. You CANNOT clear N! Ao666 without them.
Next, you have to find Inquisitor set (Malek's Armor+Nyarlaptotep's Boots) ASAP, which can
1. make you completely negates fire attacks (revenants, mancubi, archviles, nightmare archviles, cyberdemons, lava elementals - whoa!!)
2. greatly enhances your speed.
Without Inquisitor set, most characters would face a guaranteed death, but with those item (+ nanomachic laser rifle),
this map would be a pushover (if you are cautious enough to deal with their melee attacks)
Next, you need "biggest fucking nuclear gun" (nuclear BFG 9000 + BBFF, Whizkid 2) in later parts, as a subweapon.
With Son of a bitch 5 + Trigerhappy 2 + Firestorm, nanomachic laser rifle boasts its overwhelming firepower,
but it would not be enough for floor 300+ if you're get surrounded by too many bruiser brothers from start of level.
So you need it as a subweapon for critical moments.
And you would need "Shielded Armor" for nightmare demon/agony elemental caves.
(But don't forget to change your boots to cerberus boots+A and to wear Inquisitor set again before go downstairs)
And save your homing phase devices, for nightmare arachnotron caves/nightmare cacodemon caves/shambler complex.
For nightmare arachnotron/cacodemon caves, first you can do biggest fucking nuclear gun shot and deal with remaining ones with laser rifle,
but if you face shambler complex (you can know it by the opening message "The air is crackling with electricity!"),
you'd better IMMEDIATELY use homing phase device or 30~40 shamblers would teleport near you and surround you.
(Fortunately it was not generated in this run)
With overwhelming item sets, N! Ao100/ArAo666 is not difficult, actually it is easy (but really really boring)
But too bad this challenge is quite luck-dependent as you need early nano pack.
